Would it be possible to have previously drilled lug holes filled(welded) and drill out a new pattern. Say for example you had some 5 lug wheels that you wanted to use but you needed 6 lug. If so, any ideas of a place to do it.

Is it worth you life is the question you need to ask? I wouldn't do it, and I don't think any one esle would.

If the new pattern can be drilled w/o any of the new holes being dangerously close to the old holes, then it can be done.
If the position of the new holes can not be oriented this way, then you need to weigh how badly you want the new lug pattern vs. the cost of aftermarket axles. Moser makes axles in a 'stock' material that aren't as spendy as a quality Alloy axle, but aren't as strong either.

What kind of spindle or axle is it out of and what do you want to make it into? There may be other solutions to your problem...
